Welsh Premier League 2005/2006 (znana jako Vauxhall MasterFit Welsh Premier League ze względów sponsorskich) był 14. sezonem najwyższej klasy rozgrywkowej w Walii. Sezon został otwarty 27 sierpnia 2005 r., a zakończył się 30 kwietnia 2006 r. Mistrzem po raz trzeci w swojej historii został zespół Total Network Solutions.
Format rozgrywek
W sezonie 2005/2006 w lidze wystąpiło osiemnaście zespołów w systemie „każdy z każdym” (2 mecze z daną drużyną: u siebie i na wyjeździe). Drużyny w tym sezonie grały o cztery miejsca w europejskich pucharach. Jedno w pierwszej rundzie kwalifikacyjnej Ligi Mistrzów UEFA przeznaczone dla mistrza Welsh Premier League. Dwa w pierwszej rundzie kwalifikacyjnej Pucharu UEFA, z których jedno przeznaczone dla zdobywcy Pucharu Walii. Ostatnie w Pucharze Intertoto UEFA. Z ligi spadła jedna drużyna na dwie możliwe (nie spełnienie wymogów licencyjnych przez drużyny z niższego poziomu ligowego).
Skład ligi w sezonie 2005/2006
W lidze rywalizowało osiemnaście drużyn – siedemnaście z poprzedniego sezonu i jedna z Welsh Football League Division One: Cardiff Grange Harlequins (wicemistrz Welsh Football League Division One), który zastąpił Afan Lido. Z powodów licencyjnych żadna z drużyn Cymru Alliance nie otrzymała promocji, dzięki czemu w lidzie utrzymał się NEWI Cefn Druids.
